    Abstract [en]

    This report describes the investigation of how the structure of the ligand influences the product distribution in a series of model Heck reactions. The results showed that the ligand had a strong influence on the selectivity in all cases. It also describes efforts to build prediction models through PLS modelling of experimental data against calculated descriptors for the ligands in silico. In addition, a subset of the data was used to investigate the ability of DFT calculations to predict the selectivity.

    Abstract [en]

    When paper is manufactured, deposits often form in the machines. These deposits are slimelike and can interfere with the papermaking process. The slimelike deposits are aggregates of micro-organisms, also known as biofilm. One single type of micro-organism can form a biofilm, but most biofilms consists of a mixture of several different kinds of micro-organisms and can form on about any conceivable surface. To control the aggregates of micro-organisms a slimecide is added, a so-called biocide. To examine what kind of bacteria that is included in the biofilm and also which bacteria that is killed or not killed by the biocide, Terminal Restriction Fragment Length Polymorphism analysis (T-RFLP) can be used.

     

    In this report we examine biocides impact on biofilm produced in the laboratory.The biocides were first tested for possible interference with the PCR-step of the T-RFLP analysis. None of the tested ten biocides inhibited the PCR process the biofilm was formed on metal plates when these were lowered in a beaker with white water. Three different beakers were set up, one with addition of a biocide with active component 4,5-DICHLORO-1,2-DITHIOLONE from the start, one with the addition of the same biocide after three days and one with no addition at all of biocide. Samples were taken from the beakers and analyzed with T-RFLP.

     

    In this report, we show that biocides affect planktonic and biofilm micro-organisms differently. There are however some micro-organisms in the biofilm that does not get affected by the biocide.

     

    The experimental in this report is a good way of investigate the influence that biocides have on planktonic and biofilm micro-organisms, but to get even greater result the experiment should be done over a longer period of time and repeatedly.

    Abstract [en]

    A synthetic pathway towards the cap-structure of 2,2,7-trimethylguanosine containing a methylene modified triphosphate bridge have been investigated. The modification to the triphosphate bridge is hoped to slow down cap degradation and give the connected  oligunucleotide an increased lifetime. This could result in an better understanding of nuclear transport of oligonucleotides and could thereby helping to develop new treatments for different diseases. The synthesis relies on a coupling reaction between the 2,2,7-trimethylguanosine 5’phosphate and 2’-O-methyladenosine with a 5’-pyrophosphate where the central oxygen has been replaced by a methylene group. The reaction pathway consists of 9 steps of which 8 steps have been successfully performed. The last step, which includes a coupling reaction, was attempted but without successful identification and isolation of the cap-structure, and will need further attention. The reaction has been performed in a milligram scale with various yields.

    Abstract [en]

    There are few diagnostic tools for chronic musculoskeletal pain as structural imaging methods seldom reveal pathological alterations. This is especially true for Whiplash Associated Disorder, for which physical signs of persistent injuries to the neck have yet to be established. Here, we sought to visualize inflammatory processes in the neck region by means Positron Emission Tomography using the tracer (11)C-D-deprenyl, a potential marker for inflammation. Twenty-two patients with enduring pain after a rear impact car accident (Whiplash Associated Disorder grade II) and 14 healthy controls were investigated. Patients displayed significantly elevated tracer uptake in the neck, particularly in regions around the spineous process of the second cervical vertebra. This suggests that whiplash patients have signs of local persistent peripheral tissue inflammation, which may potentially serve as a diagnostic biomarker. The present investigation demonstrates that painful processes in the periphery can be objectively visualized and quantified with PET and that (11)C-D-deprenyl is a promising tracer for these purposes.

    Livsformerna på jorden delas systematiskt in i de tre domänerna bakterier, arkéer och eukaryoter. Arkéer är de mikroorganismer som lever i extrema miljöer såsom hetvattenkällor, sjöar med hög salthalt och i miljöer med extrema pH-värden. De kan existera i miljöer där inga andra organismer överlever men förekommer även rikligt överallt runtomkring oss, exempelvis i människans mage och som normalflora i munnen.

    Vissa bakterier och arkéer har genen för enzymet ammoniak monooxygenas (AMO). Detta enzym spelar en viktig roll vid rening av avloppsvatten genom att oxidera ammonium till nitrit.

    Syftet med detta examensarbete har varit att detektera arkéer i prover av aktivt slam vilket gjordes genom att optimera en Polymerase Chain Reaction (PCR) baserad metod. Först pelleterades slamproverna via centrifugering för att kunna preparera DNA. Detta DNA användes som templat för optimering av PCR med specifika primers för AMO genen hos arkéer. De PCR produkter som erhölls från det optimerade programmet klonades och transformerades in i Escherichia coli. Därefter sekvenserades PCR produkterna för att identifiera vilka ammonium oxiderande arkéer som fanns i proverna.

    De amplifierade gensekvenserna visade god överensstämmelse med den förväntade nukleotidsekvensen för arkéer. Samtliga gensekvenser passade bäst in på icke odlingsbara arkéer enligt databasen BLAST. Genom att välja bort icke odlingsbara arkéer i sökningen kunde arkéen Nitrosopumilus maritimus identifieras, vilket är en av få odlade arkéer med AMO-genen sekvens bestämd.

    Syftet med detta examensarbete var att undersöka högskolestudenters erfarenheter av övergången från studier inom kemi på gymnasial nivå till högskolenivå för att se om det finns ett glapp i undervisningen. Undersökningen bestod av dels en enkätundersökning och dels svaren från tentamensfrågor av samma karaktär som enkätfrågorna. Resultatet visar att studenterna anser att det är en skillnad mellan att läsa kemi på gymnasial nivå och på högskolenivå och att den största skillnaden ligger i ett förändrat arbetssätt. Av studenternas svar på enkätfrågorna och tentamen kan slutsatsen dras att studenterna känner till termer och begrepp, men har svårt att använda dem för att förklara kemiska fenomen.

    I detta examensarbete undersöktes hur ett kemiprov inom området kemisk jämvikt utformades och bedömdes på en gymnasieskola i Mellansverige. De två lärarna som utformade och bedömde provet intervjuades och fick svara på frågor via e- mail. Både intervjudata och frågorna via e-mail kategoriserades med hjälp av Selgheds (2006) kategorier rörande synsätt på betygsystemet. Resultatet visade att lärarna har olika synsätt på både provets utformning och bedömning trots att dem tillsammans konstruerat både prov och facit. Lärarna täckte med detta prov flera av de kända missuppfattningar som den didaktiska forskningen funnit, även om det inte var på ett systematiskt sätt. Istället visade resultatet att det fanns risk att missförstånd skall uppstå. Denna undersökning kan kompletteras med elev- och undervisningsperspektivet för att undersöka vilka konsekvenser det får om lärarna behandlar den didaktiska forskningen mer systematiskt.
